    • Gribble FM, Tucker SJ, Ashcroft FM. The essential role of the Walker A motifs of SUR1 in K-ATP channel activation by Mg-ADP and diazoxide. EMBO J 1997; 16:1145-1152. Walker A motif mutations of NBD1 and NBD2 in SUR1 do not significantly affect ATP sensitivity or channel reactivation by MgATP, but do affect MgADP potentiatory effects (activation and antagonism of ATP inhibition), suggesting the role for both NBDs in these effects. An intact NBD1 is also required for channel activation by diazoxide. Also [40].

    • + channels in the absence of the sulphonylurea receptor
    • + channels in the absence of the sulphonylurea receptor. Nature 1997; 387:179-183. Although Kir 6.2 expression does not elicit channel activity in the absence of SUR1, Kir 6.2 subunits which are truncated at the C-terminus can form functional channels when expressed alone. SUR1 does not contribute significantly to the ion permeation pathway. Significantly, these mutants exhibit intrinsic ATP sensitivity that is enhanced by coexpression with SUR1, suggesting a Kir 6.2 ATP-inhibitory site; mutation of Lys 185 results in a reduction in ATP sensitivity. SUR1 confers tolbutamide and diazoxide sensitivity on these mutant Kir 6.2 channels.

    • Morales MM, Carroll TP, Morita T, Schwiebert EM, Devuyst O, Wilson PD, et al. Both the wild type and a functional isoform of CFTR are expressed in kidney. Am J Physiol 1996; 270:F1038-F1048. CFTR is expressed predominantly in proximal tubule, TAL, and CCD. A functional alternatively spliced form that lacks transmembrane segments TM7-TM12 and NBD2, TNR-CFTR, is the primary form expressed in outer medullary collecting duct, inner medullary collecting duct, and thin limb. TNR-CFTR retains cAMP-activated Cl channel activity (inhibited by glibenclamide) and the ability to regulate outwardly rectifying Cl channels.
