Oxidative damage does not alter membrane phospholipid asymmetry in human erythrocytes
-
de Jong K, Geldwerth D, Kuypers FA: Oxidative damage does not alter membrane phospholipid asymmetry in human erythrocytes. Biochemistry 1997, 36:6768-6776. Oxidant-induced damage has been proposed to be the underlying mechanism for loss of membrane phospholipid asymmetry. This paper reports that erythrocytes did not expose PS as the result of a variety of oxidative insult, despite damage and impairment of flipase activity. The results indicate that oxidant damage does not induce exposure of phosphatidylserine on the membrane surface, but can interfere with both aminophosphilipid translocase activity or calcium-induced randomization of membrane phospholipids.

A subfamily of P-type ATPases with aminophospholipid transporting activity
-
Tang X, Halleck MS, Schlegel RA, Williamson P: A subfamily of P-type ATPases with aminophospholipid transporting activity. Science 1996, 272:1495-1497. This paper reports the cloning and sequencing of a member of subfamily of P-type ATPases with aminophosolipid transporting activity. The protein was not reported to be brought to expression to directly proof its ability to transport PS across a bilayer, but studies in Saccharomyces cerevisiae suggest that aminophospholipid translocation is a general function of members of this protein family.

Production and characterization of a mutant cell line defective in aminophospholipid translocase
-
Zhao J, Sims PJ, Wiedmer T: Production and characterization of a mutant cell line defective in aminophospholipid translocase. Biochim Biophys Acta 1997, 1357:57-64. The identification of cell lines that have a decreased flipase activity as reported in this paper, will be powerful tools, suitable for expression cloning of cDNAs coding for candidate flipases. Similar approaches will be important to characterize other proteins involved in maintenance of membrane phospholipid organization.

Test ST, Mitsuyoshi J: Activation of the alternative pathway of complement by calcium-loaded erythrocytes resulting from loss of membrane phospholipid asymmetry. J Lab Clin Med 1997, 130:169-182. The importance of PS exposure is well established in regulatory mechanisms of hemostasis. This study demonstrates that loss of phospholipid asymmetry in erythrocytes not only results in expression of procoagulant activity but also renders the cells capable of affecting the alternative pathway of complement.

Appearance of phosphatidylserine on apoptotic cells requires calcium-mediated nonspecific flip-flop and is enhanced by loss of the aminophospholipid translocase
-
Bratton DL, Fadok VA, Richter DA, Kailey JM, Guthrie LA, Henson PM: Appearance of phosphatidylserine on apoptotic cells requires calcium-mediated nonspecific flip-flop and is enhanced by loss of the aminophospholipid translocase. J Biol Chem 1997, 272:26159-26165. In the last few years, it has become apparent that PS exposure is an important early step in apoptosis. This paper describes aspects of the underlying mechanisms that determine this loss of phospholipid asymmetry, and identifies calcium as a key factor.
