Relation of common allelic variation at vitamin D receptor locus to bone mineral density and postmenopausal bone loss: Cross-sectional and longitudinal population study

Jørgensen HL, Hassager C, Sand JC, Bjuring M, Christiansen C: Relation •• of common allelic variation at vitamin D receptor locus to bone mineral density and postmenopausal bone loss: cross-sectional and longitudinal population study. BMJ 1996, 313:586-590. In this relatively large study of approximately 900 Danish women stratified by age in pre-, peri-, and postmenopausal groups, no correlation was found between VDR genotype and bone density (spine, proximal femur, and forearm) in any age groups. No correlation with bone loss calculated by BMD measurements was found over an 18-year period.

The Bsml vitamin D receptor restriction fragment length polymorphism (bb) influences the effect of calcium intake on bone mineral density
-
Kiel DP, Myers RH, Cupples LA Kong XF, Zhu XH, Ordovas J, Schaeffer •• EJ, Felson DT, Rush D, Wilson PWF, Eisman JA, Holick MF: The Bsml vitamin D receptor restriction fragment length polymorphism (bb) influences the effect of calcium intake on bone mineral density. J Bone Miner Res 1997, 12:1049-1057. Cross-sectional data from the Framingham study cohort demonstrates that the VDR genotype affects the interaction of dietary calcium with bone density. Association between calcium intake and bone density was found only in the bb genotype. Differences in BMD were observed at high (800 mg/d) intakes, a result at variance with other studies.

Zmuda JM, Cauley JA, Danielson ME, Wolf RL, Ferrel RE: Vitamin D • receptor gene polymorphisms, bone turnover, and rates of bone loss in older African-American women. J Bone Miner Res 1997, 12:1446-1452. No association between VDR polymorphism and biochemical markers of bone turnover in 101 elderly African-American women, although the tt genotype group lost bone at a faster rate than the TT group. Also, women homozygous for the B allele exhibited a lower fractional calcium absorption.

Vitamin D receptor gene polymorphism in men and its effect on bone density and calcium absorption
-
Francis RM, Harrington F, Turner E, Papiha SS, Datta HK: Vitamin D • receptor gene polymorphism in men and its effect on bone density and calcium absorption. Clin Endocrinol 1997, 46:83-86. Study on a small series of men, showing no association between VDR polymorphism and either bone density (lumbar spine and proximal femur), or fractional calcium absorption.

The effect of vitamin D supplementation on the bone mineral density of the femoral neck is associated with vitamin D receptor genotype
-
Graafmans WC, Lips P, Ooms MAC, Van Leeuwen JPTM, Pols HAP, • Uitterlinden AG: The effect of vitamin D supplementation on the bone mineral density of the femoral neck is associated with vitamin D receptor genotype. J Bone Miner Res 1997, 12:1241-1245. Small size study (81 elderly women) demonstrating response to vitamin D supplementation (400 IU daily for at least 2 years) results in increases in femoral neck BMD in both BB and Bb genotypes, but not in the bb variant, despite no differences in BMD at baseline.
