The authors studied a mouse heterozygous for X-linked ornithine transcarbamylase expression; three-dimensional reconstruction revealed no definite orientation of patches such as along the portal-central vein axis.

This is a particularly significant experiment in the light of future prospects for ex vivo gene therapy, since the authors showed it was possible to genetically modify isolated oval cells (expressed β-galactosidase after retroviral infection) and return them in vivo where they functioned as hepatocytes. The recipient rats were initially analbuminaemic but, after oval cell transplantation, serum albumin was elevated for at least ten weeks and the tagged oval cells differentiated into albumin-expressing hepatocytes.

Thus, a certain proportion of hepatocytes have a replicative potential on a par with haemopoietic stem cells.
