Mice lacking serum paraoxonase are susceptible to organophosphate toxicity and atherosclerosis
-
In press
-
Shih DM, Gu L, Xia Y-R, Navab M, Li W-F, Hama S, et al. Mice lacking serum paraoxonase are susceptible to organophosphate toxicity and atherosclerosis. Nature 1998 In press. The first description of transgenically modified mice which lack PON1. These mice are at increased susceptibility to organophosphate poisoning and atherosclerosis indicating a central role for PON1 in vivo in protecting against both. This is particularly important for the antiatherogenic properties of PON1 which had previously only been shown in in-vitro systems.

Effect of the human serum paraoxonase 55 and 192 genetic polymorphisms on the protection by high density lipoprotein against low density lipoprotein oxidative modification. FEBS Letts 1998; 423:57-60. This manuscript describes how the presence of the different PON1 isoenzymes governs the ability of HDL to protect LDL against oxidative modification. This is apparently due to the differential rates of hydrolysis ot lipid peroxides by the PON1 isoenzymes.
