Analysis of Hoxd13 and Hoxd11 misexpression in chick limb buds reveals that Hox genes affect both bone condensation and growth
-
Goff D, Tabin C: Analysis of Hoxd13 and Hoxd11 misexpression in chick limb buds reveals that Hox genes affect both bone condensation and growth. Development 1997, 124:627-636. By misexpression of Hoxd11 and Hoxd13 using viral vectors in chick limbs, it was shown that Hox genes may act to pattern skeletal structures during early mesenchymal/cartilage condensations or later by affecting cell division within the proliferative zone of growing cartilage.

-
Muragaki Y, Mundlos S, Upton J, Olsen BR: Altered growth and branching patterns in synpolydactyly caused by mutations in HOXD13. Science 1996, 272:548-551. Mutation of HOXA13 in hand-foot-genital syndrome
-
Mortlock DP, Innis JW: Mutation of HOXA13 in hand-foot-genital syndrome. Nature Genetics 1997, 15:179-180. The authors reported the identification of a mutation in the human HOXA13 gene in the originally reported family (Stem et al. J Pediatr 1970, 77:109-116.) with HFGS. The skeletal phenotype of the hypodactyly mice reported by Mortlock et al. (Nature Genetics 1996, 13:284-289.) closely resembled the limb anomalies reported in HFGS and were a clue to the potential mutant gene in this human syndrome.

Synpolydactyly phenotypes correlate with size of expansions in HOXD13 polyalanine tract
-
Goodman F, Mundlos S, Muragaki Y, Donnai D, Giovannucci-Uzielli M, Lapi E, Majewski F, McGaughran J, McKeown C, Reardon W, Upton J, Winter R, Olsen B, Scambler P: Synpolydactyly phenotypes correlate with size of expansions in HOXD13 polyalanine tract. Proc Natl Acad Sci U S A 1997, 94:7458-7463. Sixteen new families with synpolydactyly were evaluated at the molecular level for HOXD13 polyalanine tract expansions. Increasing the number of alanines in the expansion increases the number of affected limbs and the severity of the malformations. The only example in which a genitourinary defect could be associated with synpolydactyly was found in a single family who had the largest polyalanine expansion (14 residues) ever observed.

Synpolydactyly in mice with a targeted deficiency in the HoxD complex
-
Zakany J, Duboule D: Synpolydactyly in mice with a targeted deficiency in the HoxD complex. Nature 1996, 384:69-71. The authors engineered a deficiency of Hoxd13, Hoxd12, and Hoxd11 simultaneously in mice. The limbs of these mice resemble those of humans with synpolydactyly, suggesting that the human synpolydactyly phenotype may be due to a negative effect of the HOXD13 alanine duplication on the expression of other HOXD genes.
