By misexpression of Hoxd11 and Hoxd13 using viral vectors in chick limbs, it was shown that Hox genes may act to pattern skeletal structures during early mesenchymal/cartilage condensations or later by affecting cell division within the proliferative zone of growing cartilage.

Proc Natl Acad Sci U S A 1997, 94:7458-7463. Sixteen new families with synpolydactyly were evaluated at the molecular level for HOXD13 polyalanine tract expansions. Increasing the number of alanines in the expansion increases the number of affected limbs and the severity of the malformations. The only example in which a genitourinary defect could be associated with synpolydactyly was found in a single family who had the largest polyalanine expansion (14 residues) ever observed.

The authors engineered a deficiency of Hoxd13, Hoxd12, and Hoxd11 simultaneously in mice. The limbs of these mice resemble those of humans with synpolydactyly, suggesting that the human synpolydactyly phenotype may be due to a negative effect of the HOXD13 alanine duplication on the expression of other HOXD genes.
