Cathepsin K but not cathepsin B, L or S is abundantly expressed in human osteoclasts
-
Drake FH, Dodds R, James I, Connor J, Debouck C, Richardson S, Lee-Rykaczewski E, Rieman D, Barthlow R, Hastings G, Gowen M: Cathepsin K but not cathepsin B, L or S is abundantly expressed in human osteoclasts. J Biol Chem 1996, 271:12511-12516. The authors analyzed the contents of a cDNA library generated from human osteoclast cells. Cathepsin K, a putative cysteine protease, was isolated and was subsequently shown [19] to be involved in bone resorption in vivo. Without a cDNA library, it is unlikely that this protein would have been isolated. Research efforts are focusing on developing inhibitors of this protease for treatment of osteoporosis.

Small peptides as potent mimetics of the protein hormone erythropoietin
-
Wrighton NC, Farrell FX, Chang R, Kashyap AK, Barbone FP, Mulcahy LS, Johnson DL, Barrett RW, Jolliffe LK, Dower WJ: Small peptides as potent mimetics of the protein hormone erythropoietin. Science 1996, 273:458-463. The authors report the isolation of a small (20-residue peptide) agonist at the erythropoietin (EPO) receptor by screening a combinatorial phage display library. Natural ligands of EPO are large folded proteins, and searches for smaller ligands have been unfruitful until now.
