-
Lynch KW, Maniatis T: Assembly of specific SR protein complexes on distinct regulatory elements of the Drosophila doublesex splicing enhancer. Genes Dev 1996, 10:2089-2101. A detailed study of protein binding on Drosophila doublesex splicing enhancer. Splicing factors Transformer, Transformer 2 are shown to recruit different SR proteins to the repeat region and the purine-rich element of the doublesex splicing enhancer. The binding of Transformer, Transformer 2 and SR proteins to the RNA is synergistic. The relative positions of these bound splicing factors on the RNA were determined.

Min H, Turck CW, Black DL: A new regulatory protein, KSR, mediates exon inclusion through an intronic splicing enhancer. Genes Dev 1997, in press. By using neural-specific inclusion of c-src N1 exon as a working model, these authors have identified and characterized a 75 kDa protein, KSR, required for N1 splicing in vitro. KSR contains KH-type RNA binding motifs and may play an important role in neural-specific splicing.

SR proteins that are essential for generic pre-mRNA splicing inhibit splicing of a regulated adenovirus pre-mRNA
-
Kanopka A Mühlemann O, Akusjärvi G: SR proteins that are essential for generic pre-mRNA splicing inhibit splicing of a regulated adenovirus pre-mRNA. Nature 1995, 381:535-538. An intronic sequence element is shown to repress splicing of adenovirus IIIa mRNA splicing. SR proteins, specifically ASF/SF2, are found to bind the element and to mediate splicing inhibition, probably by steric hindrance. The position of the element in the pre-mRNA determines whether its effect is either positive or negative.

-
Adams MD, Tarng RS, Rio DC: The alternative splicing factor PSI regulates P-element third intron splicing in vivo. Genes Dev 1997, 11:129-138. Experiments carried out in Drosophila demonstrate that PSI is a tissue specific splicing inhibitor in vivo. Ectopic expression of PSI in the female germline inhibits P element IVS3 silencing and blocking expression of PSI results in activation of IVS3 splicing in soma.

The Clk/Sty protein kinase phosphorylates SR splicing factors and regulates their intracellular distribution
-
Colwill K, Pawson T, Andrews B, Prasad J, Manley JL, Bell JC, Duncan PI: The Clk/Sty protein kinase phosphorylates SR splicing factors and regulates their intracellular distribution. EMBO J 1996, 15:265-275. Clk/Sty kinase is shown to interact with SR proteins in yeast two-hybrid screening. In vitro assays indicate that Clk/Sty phosphorylates the RS domain of ASF/SF2 in a manner very similar to that detected in vivo. Clk/Sty colocalizes with SR proteins in vivo.

Sequence-specific RNA binding by an SR protein requires RS domain phosphorylation: Creation of an SRp40-specific splicing enhancer
-
Tacke R, Chen Y, Manley JL: Sequence-specific RNA binding by an SR protein requires RS domain phosphorylation: creation of an SRp40-specific splicing enhancer. Proc Natl Acad Sci USA 1997, 94:1146-1153. This study shows that phosphorylation of the RS domain of SRp40 is essential for sequence-specific RNA binding. High-affinity binding sites for SRp40 selected by SELEX function as splicing enhancers in vitro.

Phosphorylation of the ASF/SF2 RS domain affects both protein-protein and protein-RNA interactions and is necessary for splicing
-
Xiao SH, Manley JL: Phosphorylation of the ASF/SF2 RS domain affects both protein-protein and protein-RNA interactions and is necessary for splicing. Genes Dev 1997, 11:334-344. Phosphorylation of ASF/SF2 enhances an RS domain-mediated interaction with U1 70K protein and with U1 snRNP. Conditions are described in which phosphorylation of ASF/SF2 is found to be necessary for efficient splicing in vitro.
