Using a genetic suppression assay, in mammalian tissue culture, a novel intermolecular helix (heitx III) between U2 and U6 is identified. Evidence that this helix helps to juxtapose the 5′ splice site and the branch site is provided by suppression of a branch-site mutation by a network of compensatory changes in the 5′ splice site and in the U2 and U6 snRNAs.
