J Clin Invest 1996, 97:154-161. In CETP transgenic mice mated with apo E or LDL receptor gene knockout mice, plasma CETP and hepatic CETP mRNA levels were markedly increased after cholesterol feeding. In CETP transgenic, CETP transgenic/apo E knockout, and CETP transgenic/LDL receptor knockout mice on different diets, plasma CETP and CETP mRNA levels were correlated with plasma cholesterol levels. Hepatic CETP gene expression may be controlled by a novel mechanism that senses changes in plasma cholesterol levels independent of apo E and LDL receptor levels.

Arterioscler Thromb Vasc Biol 1995, 15:1849-1856. This study was the first demonstration of atherogenicity in CETP-deficient individuals. Atherosclerotic cardiovascular diseases were observed in CETP-deficient individuals despite extremely high HDL-cholesterol levels. These occur especially in those patients with a combined deficiency of CETP and HTGL, which have a synergistic role in the formation of small HDL particles.

Eur J Clin Invest 1995, 25:332-339. In this report, the apo B-containing triglyceride-rich LDL particles from CETP-deficient individuals are shown to have a lower affinity to LDL receptors than those from control individuals. Thus, CETP deficiency causes functional abnormalities of both HDL and LDL as well as compositional abnormalities of these Lps.
