    • Perturbation of nuclear architecture by long-distance chromosome interactions
    • Dominant heterochromatic insertion located distally on chromosome arm 2R of Drosophila was found to frequently associate with the pericentric heterochromatin of 2R, in support of a nuclear compartmentalization model for trans-inactivation of the brown gene. Significant associations were observed in various larval tissues, but not in early embryos before the onset of pairing and heterochromatin formation.

    • Dominant insertion and heterochromatin is reduced by chromosome rearrangement and when Suppressors-of-variegation are present, in correlation with trans-inactivation of brown. The imprinted region of human chromosome 15 that includes the Prader-Willi and Angelmann syndrome loci displays homologous associations during late S phase of the cell cycle. Association was deficient in patients affected with these syndromes because they carried two copies of the region from one parent, suggesting that homologous pairing of oppositely imprinted chromosome domains is involved in normal imprinting.

The use of site-specific recombination permitted the measurement of homologs pairing during development. Structural heterozygosity caused by proximal breaks in chromosomes reduced recombination at a site, supporting the pairing interpretation of transvection effects. It is suggested that longer cell cycle times allow homologs to zipper up, which could explain why some pairing-dependent interactions are insensitive to disruption by proximal breaks.
