This transgenic study, performed in ES cells and mouse chimeras, demonstrated that a 450 kb YAC transgene derived from the Xic enabled cells to count, choose, and induce Xist expression from both the X- and transgene-containing autosomes, thus arguing that the 450 kb transgene is sufficient for initiation of X inactivation. A marker gene carried on the transgene could be silenced in cis to Xic sequences.

A targeted deletion of Xist encompassing the promoter and exon 1 was created in female ES cells and demonstrated definitively that Xist is required for X inactivatiori in cis. As the Xist deletion did not disrupt X inactivation on the wild-type chromosome, this study suggests that chromosome-counting must be achieved by a Xist-independent mechanism.

When a transgenic line with a 100 kb YAC deletion was integrated into the Y, however, Xist was observed at high levels comparable to that seen in somatic female cells.
