This nice paper is the first to measure the actual affinities involved in the reaction between TCRs and ligands which can positively select thymocytes or activate mature T cells bearing these TCRs. The paper should be read with the caveat that these phenomena are probably not controlled by affinity alone. The avidity with which the cells interact, which will be affected by ligand and TCR concentration and interactions between other surface molecules, is probably the crucial factor.

of special interest. This paper took advantage of the production of mice in which all MHC class II proteins were bound to a single peptide. Therefore all the class II-restricted T cells in the mouse must have been positively selected on that combination. Nevertheless their responses as mature cells were not antagonized by the selecting ligand.

+ T cells appear in these mice, demonstrating that one MHC-peptide combination can positively select many thymocytes. Most of the selected cells react with the same MHC class II protein bound to the many self peptides which can engage it in normal mice. Thus most of the T cells which are positively selected by the single MHC-peptide combination would normally be clonally deleted in wild type mice.
