Eur J Cancer 1996, 32A:1179-1184. This thorough clinical investigation examines the hypothesis that individual variations in systemic ifosfamide metabolism are associated with a higher risk of nephropathy. Results show a negative correlation between production of dechloroethylated ifosfamide metabolites and renal injury. The authors speculate that differences in renal rather than hepatic drug metabolism may be responsible for nephrotoxicity.

Am J Obstet Gynecol 1996, 175:270-274. This study convincingly demonstrates that inhibition of γ-glutamyl transpeptidase prevents cisplatin nephrotoxicity without altering kidney platinum uptake or content. It is an excellent example of potential glutathione conjugate-mediated toxicity and of the importance of considering renal metabolism when evaluating drug-induced nephrotoxicity.

Glycoprotein 330 is concentrated in the apical region of proximal tubule cells. It was initially described as a major target antigen in Heymann nephritis, an experimental model of antibody-mediated membranous glomerulonephritis. It now appears that this glycoprotein is a drug receptor that mediates cell uptake and toxicity of aminoglycoside.
