In this study, the authors isolated a previously unrecognized surface-exposed 28 000 M, heparin-binding hemagglutinin from M. bows BCG and H37Ra M. tuberculosis, Monoclonal antibodies that recognize the protein demonstrate that it is present on the surface of H37Ra M. tuberculosis. This protein mediates mycobacterial aggregation and the adherence of mycobacteria to Chinese hamster ovary cells.

The authors provide evidence for the activation of PLD during phagocytosis of the Erdman and H37Ra strains of M. tuberculosis by MDMs. Inhibition of PLD activity results in reduced phagocytosis of bacteria, indicating that these processes may be linked and that activation of PLD may regulate bacterial ingestion, M. tuberculosis phagocytosis also results in rapid and sustained increases in tyrosine phosphorylation of multiple MDM proteins, Protein tyrosine kinases may thus be involved in the PLD activity.

Using cryosection immunogold labelling techniques, the authors demonstrated that phagosomes containing live M. tuberculosis acquire exogenously administered transferrin, a marker for early endosomes. This study shows that the M. tuberculosis phagosome can fuse with early endosomes, providing a potential targeted approach to antimicrobial therapy.

This study indicates that the M. tuberculosis phagosome can acquire markers from the cell surface, and that this pathway is distinct from that of fluid-phase markers.

The authors provide evidence that infection of a murine bone marrow macrophage clone with live virulent M. tuberculosis allows for the specific recognition of soluble ovalbumin epitopes by a MHC class I-restricted ovalbumin-specific cytotoxic T-lymphocyte hybridoma. This study thus indicates that M. tuberculosis provides access of a soluble antigen from the endocytic compartment to the macrophage MHC class I pathway. All of the MHC class I presentation of albumin facilitated by M. tuberculosisinfected macrophages was TAP-dependent.

The authors provide evidence that mannose-capped LAM from M. tuberculosis as well as AraLAM could induce T-cell migration in the absence of serum using a modified Boyden chamber assay. In contrast, mannose-capped LAM from M. bovis BCG failed to induce this effect. Chemotaxis was dependent upon the acyl groups of LAM, new protein synthesis and protein tyrosine kinases. Experiments using human alveolar macrophages indicated that LAM may be one of the chemotactic factors released by M. tuberculosis-infected macrophages.
