Science 1995, 270:1677-1680. Shows, for the first time, the in vivo importance of editing the Q/R site in AMPA receptor subunit GluR-B pre-mRNA. Mice were constructed to carry one GluR-B allele that was incompetent for Q/R site editing and one wild-type allele. These mice developed a severe epileptic syndrome and died within three weeks of birth.

This study localizes the intronic ECS for Q/R site editing in GluR5 and GluR6 pre-mRNA. This cis-acting element is approximately 2000 nucleotides 3′ of the exonic editing site. Although long-range interactions in RNA are common, the results of this study imply a large range of distances between exonic and intronic site and intronic ECS in RNAs edited by adenosine deamination.
