-
Cheng D, Chang CC, Qu X, Chang TY: Activation of acyl-coenzyme A: cholesterol acyltransferase by cholesterol or by oxysterol in a cell-free system. J Biol Chem 1995, 270:685-695. High level expression of the hACAT cDNA in insect cells, followed by partial protein purification and liposome-mediated resolubilization, demonstrates the kinetic and regulatory profiles of the human enzyme. In addition, cholesterol and oxysterol are demonstrated to be allosteric effectors of ACAT regulation.

Sterol esterification in yeast: A two gene process
-
Yang H, Bard M, Bruner DA, Gleeson A, Deckelbaum RJ, Aljinovic G, Pohl T, Rothstein R, Sturley SL: Sterol esterification in yeast: a two gene process. Science 1996, 272:1353-1356. The authors identified a gene family in this model eukaryote that is responsible for sterol esterification. Deletion of both genes nullifies sterol esterification and down-regulates sterol biosynthesis. The concept of multiple gene families for ACAT in all organisms is proposed based on the isolation of novel ACAT homologs from the database of expressed sequence tags.

Proc Natl Acad Sci U S A 1996, 93:14041-14046. The authors present the first mutant animal model for the ACAT reaction and illustrate the effect of loss of activity on intracellular sterol accumulation and serum cholesterol levels. Importantly, a tissue-specific profile of activity is demonstrated in the knockout animal, clearly affirming the concept of multiple genes for the process.
