The authors stably transfected a cell line with the vaccinia virus uracil DNA glycoslylase gene, encoding an enzyme required for viral DNA replication. The cells were then used to isolate and propagate a mutant vaccinia virus with a deleted uracil DNA glycosylase gene. In non-permissive cells, prolonged early gene expression was detected, suggesting the potential of the mutant virus as an expression vector.
