Incubation of purified ribosome-nascent chain complexes with SRP or SRP subcomplexes shows that the binding of GTP to SRP54 is induced by the interaction of SRP with the ribosome. Subsequent hydrolysis of GTP is induced by the interaction of the complex of ribosome, nascent chain, and SRP with the SRP receptor. Together with the results in [12,14•], these results lead to a scheme of how GTP regulates the first steps in translocation across the ER membrane (see also Fig. 1).

The mutual regulation of the activity of the GTPases involved in the SRP pathway was studied with a model system employing bacterial homologs of SRP54 and SRα (Ffh and FtsY, respectively). The nucleotide specificity of FtsY was altered from GTP to XTP by mutagenesis. FtsY in the XTP-bound form stimulated GTP hydrolysis by Ffh, and its own XTP hydrolysis required Ffh in the GTP-bound form. Thus, the two GTPases seem to act on each other as GAPs.

The insertion of nascent polypeptides into the translocation site of the mammalian ER membrane is shown to occur in two steps: first, binding of the translating ribosome to the membrane, and second, productive insertion of the nascent chain into the translocation channel. The latter step requires a critical length of the nascent chain, in addition to a functional signal sequence. In case of the model protein preprolactin, the Sec61p complex in the lipid bilayer is sufficient for nascent chain insertion and signal sequence recognition.

A model of how the NAC and SRP cooperatively regulate signal sequence specific targeting is proposed.

The hydrophobic cores of both sequences may be cross-linked to phospholipids, whereas the carboxy-terminal hydrophilic region is in the vicinity of Sec61p. Thus, a portion of the nascent chain located in the translocation channel has contact with the surrounding lipid phase, suggesting that the channel is laterally open early during translocalion.

In US11-expressing cells, MHC1 molecules are inserted into the ER and core glycosylated. With a half-life of 1 minute, however, they are dislocated to the cytosol, deglycosylated and targeted to a degradation pathway involving the proteasome.

The complex consisted of two subcomplexes: the Sec61p complex which also exists as a separate entity, and the Sec62/63p complex. When reconstituted into proteoliposomes, the heptameric complex, but neither of the individual subcomplexes, promoted posttranslational translocation. Transport was stimulated by Kar2p in an ATP-dependent manner. For the model protein prepro-α-factor, a base level of transport was observed even in the absence of Kar2p and ATP.
