Parental occurrence of stroke and risk of stroke in their children: the Framingham study

This is a detailed, community-based prospective study that demonstrated parental stroke by 65 years of age was associated with a threefold increase in risk of offspring stroke, even when adjusting for conventional stroke risk factors. As such, verified parental stroke may serve as a clinically useful risk marker of an individual's propensity to stroke

Sequence variants on chromosome 9p21.3 confer risk for atherosclerotic stroke
-
This study demonstrated that the chromosome 9p21.3 region, which had previously been associated with coronary artery disease, also represents a major risk locus for atherosclerotic stroke. The study also demonstrated that the effect of this locus on stroke appears to be independent of its relation to coronary artery disease and other stroke risk factors

The Causative classification of stroke system: an international reliability and optimization study
-
This article describes how a new stroke classification system, the CCS, allows classification of stroke subtypes by multiple investigators with high reliability, thereby supporting its potential for improving stroke classification in multicenter studies. The CCS is hoped to alleviate some of the ambiguities generated by the widely-employed TOAST classification system

Dabigatran versus warfarin in patients with atrial fibrillation
-
Although not a genetics study, this large randomized study demonstrated that in patients with atrial fibrillation, dabigatran as compared with warfarin was associated with lower rates of stroke and systemic embolism. On the basis of this trial, dabigatran is now approved for stroke prevention in the setting of atrial fibrillation in the United States
