    • Htm1 protein generates the N-glycan signal for glycoprotein degradation in the endoplasmic reticulum
    • Using S. cerevisiae mutants, this paper shows that the exposure of an α1,6 linked mannose residue, through the activity of Htm1 (EDEM1) is essential for ERAD. It modifies the previous notion that removal of mannose-b from M9 to produce M8B is sufficient for targeting of a glycoprotein to ERAD in yeast.

    • Defining the glycan destruction signal for endoplasmic reticulum-associated degradation
    • This paper describes the specificity of the S. cerevisiae lectin Yos9 in vitro, which strengthens its proposed role as a receptor for trimmed N-glycans that targets misfolded glycoproteins to ERAD and suggests that in yeast, as in mammalian cells, there is trimming to M5-6 to target a glycoprotein for ERAD. • This paper shows that the activity of ERManI is required for trimming of N-glycans to M5-6 as a signal for targeting glycoproteins to ERAD in mammalian cells. • This paper determines the specificity of human OS9 in vitro, which strengthens its proposed role as a receptor for trimmed N-glycans that targets misfolded glycoproteins to ERAD. It shows that removal of mannose-c and exposure of an α1,6 linked mannose residue is the feature recognized by OS9.
