Hussey Arabidopsis homologues of the autophagy protein Atg8 are a novel family of microtubule binding proteins FEBS Lett 567 2004 302 306 The authors show that principal components of the autophagy pathway found in animals and fungi are conserved in plants, with the Arabidopsis homologues Atg4 and Atg8 able to partially complement yeast knockouts of the equivalent genes. Atg8 was also shown to bind MTs, thereby linking the cytoskeleton to autophagy (the non-selective degradation of cytosolic material and superfluous organelles occur in the lysosome or vacuole).

    • S.L. Shaw, R. Kamyar, and D.W. Ehrhardt Sustained microtubule treadmilling in Arabidopsis cortical arrays Science 300 2003 1715 1718 The authors used time-lapsed imaging to study the dynamics of MTs labeled with GFP-TUBULIN in Arabidopsis seedlings, revealing that interphase MTs originate de novo along the cell cortex. These MTs exhibited rapid growth and shrinkage at their plus ends and slow, intermittent depolymerization at their minus ends. This "hybrid treadmilling" drives MT repositioning and the reorganization of cytoskeletal arrays.

    • J. Chan, G.M. Calder, J.H. Doonan, and C.W. Lloyd EB1 reveals mobile microtubule nucleation sites in Arabidopsis Nat Cell Biol 5 2003 967 971 The authors show that two Arabidopsis EB1-like proteins fused to GFP localize to MT plus ends as well as to presumed cortical MT nucleation sites. Hülskamp A novel localization pattern for an EB1-like protein links microtubule dynamics to endomembrane organization Curr Biol 13 2003 1991 1997 The authors show that GFP-AtEB1 expressed in Arabidopsis localizes to MT plus ends as well as around organelles and endomembranes that frequently merge with the endoplasmic reticulum. Additionally, localization data suggested that AtEB1 behaves differently than other EB1 proteins in that it localizes to already-stabilized MTs instead of acting as a MT stabilizer.

    • J.C. Sedbrook, D.W. Ehrhardt, S.E. Fisher, W.R. Scheible, and C.R. Somerville The Arabidopsis SKU6/SPIRAL1 gene encodes a plus end-localized microtubule-interacting protein involved in directional cell expansion Plant Cell 16 2004 1506 1520 The authors show that a null mutation in the SPR1 gene affects anisotropic cell expansion in complex ways, making plant organs twist about their axes and roots respond abnormally to touch stimulation. SPR1 was found to encode a novel plant-specific protein that localized to MT plus ends as they grew, but fell off upon depolymerization. The authors also present data that show that the parallel orientation of cortical MTs in cells in the root elongation zone did not always correlate with the direction of cell expansion, thus questioning a model in which cortical MT pitch drives directional cell expansion.

NtMAP65-1b could not bind to subtilisin-digested tubulin, presumably because subtilisin removed the exposed carboxy terminus of tubulin. • The plant microtubule-associated protein AtMAP65-3/PLE is essential for cytokinetic phragmoplast function
    • S. Muller, A. Smertenko, V. Wagner, M. Heinrich, P.J. Hussey, and M.T. Hauser The plant microtubule-associated protein AtMAP65-3/PLE is essential for cytokinetic phragmoplast function Curr Biol 14 2004 412 417 This study provides evidence that the Arabidopsis protein AtMAP65-3 is essential for the maintenance of normal phragmoplast shape and function because AtMAP65-3 mutants displayed aberrant cytokinesis and mishappen phragmoplasts. An antibody that is specific to AtMAP65-3 located only to the mitotic arrays, including the phragmoplast midline, where AtMAP65-3 might function by cross-linking overlapping MT plus ends.
