Nanotubular highways for intercellular organelle transport
-
This study describes for the first time tunneling nanotubes (TNT). Their characterization by scanning electron microscope revealed a seamless transition between the membrane of the nanotube and the plasma membrane of the connected cells. In addition, transmission electron microscopy showed that the membrane of TNT was continuous with the membrane of the connected cells. By fluorescence microscopy, the intercellular transfer of organelles of endocytic origin, actin and, to a limited extent membrane components was observed.

Functional connectivity between immune cells mediated by tunneling nanotubules

This represents the first report on TNT-mediated functional connectivity. It was shown that calcium signals, induced in myeloid cells, were propagated via TNT to neighboring cells, where they elicited a physiological reaction. In addition to calcium, microinjected dye tracers could be transferred through these connections.

This study shows that membrane nanotubes connecting T cells are close-ended bridges. A junctional border was observed between the nanotube and the connected cell at the ultrastructural level, the membrane components did not seamlessly mix along the length of the nanotube and calcium signals were not propagated between nanotube-connected cells. Furthermore they show that innate membrane nanotubes provide a new route for HIV-1 transmission between T cells in a receptor-mediated manner.

This paper reports for the first time on the TNT-mediated unidirectional transfer of mitochondria from cardiomyocytes to endothelial stem cells as well as the transfer of cytoplasmically expressed GFP. It is of note that concurrent with the transfer of mitochondria, transdifferentiation of the endothelial progeniator cells into cardiomyocytes was observed.
