TNTs permit membrane traffic of vesicles between networks of cells, suggesting an alternative mechanism of organelle transfer between cells by membrane continuity, rather than classical exo-and endocytosis. TNTs do not permit cytoplasmic transport, however. Like PD, trafficking along TNTs is sensitive to latrunculin B, and does not involve microtubules. The discovery promises to open up novel lines of research at the intersection of plant and animal cell biology.

The authors show that KN1 cannot move in mature leaves from the epidermis into mesophyll, but can indeed move from the epidermal layer into inner tissues in the meristem. This observation supports the idea that the PD maturation process and PD closure to certain developmental signals is coupled to cell differentiation. The causative arrow is not clear though, and future work using KN1 as a tool to dissect symplasmic isolation will give new insights and evidence.

Wu et al. [37] show data using LFY∼GFP deletions that movement of LFY in the meristem is consistent with diffusion. However Lee et al. have identified a cell factor that may affect LFY trafficking in the meristem. These groups should design experiments together to test the specific way in which NCAPP1 affects LFY trafficking.
