This is an extensive yeast-two hybrid analysis that identified a large number of proteins that interact with single PDZ domains of PDZK1. These associated proteins can be divided into transporters [NaPi-II, NaPi-I, NHE3, OCT-N1, CFEX (mouse orthologue of human solute carrier family 26), URAT1 (urate transporter 1)], membrane-associated proteins (MAP17) and regulatory factors [NHERF1, DAKAP2 (dual kinase A anchoring protein 2)]. All interacting proteins immunolocalize in proximal tubules.

J Biol Chem 2004; 279:19051-19063. This study demonstrates that a complex containing SAP (synapse-associated protein) 97, CASK, Veli, and Mint1 associates with the C-terminal PDZ-binding domains of Kir2 channels. The authors propose two association models. In one, SAP97 would interact with Kir2 channels and recruit CASK, Veli, and Mint; in the alternative model, Veli would interact with Kir2 channels and and recruit CASK and SAP97.

This study shows that pharmacological activation of PKA by cAMP inhibits NHE3 activity in BBM isolated from wild-type mice, but has no effect on samples from NHERF1-knockout animals. This lack of response is associated with a reduced cAMP-induced phosphorylation of NHE3.

This paper describes how phosphorylation by PKC of the PDZ2 domain NHERF1 (on Ser-162) decreases the ability of NHERFI to bind CFTR. In addition, a mutation in NHERF1 that mimics phosphorylation (Ser-162 to Asp) decreases CFTR binding and disrupts the ability of NHERF to stimulate CFTR channel activity.
