PDZK1: I. A major scaffolder in brush borders of proximal tubular cells
-
Gisler SM, Pribanic S, Bacic D, et al. PDZK1: I. a major scaffolder in brush borders of proximal tubular cells. Kidney Int 2003; 64:1733-1745. This is an extensive yeast-two hybrid analysis that identified a large number of proteins that interact with single PDZ domains of PDZK1. These associated proteins can be divided into transporters [NaPi-II, NaPi-I, NHE3, OCT-N1, CFEX (mouse orthologue of human solute carrier family 26), URAT1 (urate transporter 1)], membrane-associated proteins (MAP17) and regulatory factors [NHERF1, DAKAP2 (dual kinase A anchoring protein 2)]. All interacting proteins immunolocalize in proximal tubules.

A multiprotein trafficking complex composed of SAP97, CASK, Veli and Mint1 is associated with inward rectifier Kir2 potassium channels
-
Leonoudakis D, Conti LR, Radeke CM, et al. A multiprotein trafficking complex composed of SAP97, CASK, Veli and Mint1 is associated with inward rectifier Kir2 potassium channels. J Biol Chem 2004; 279:19051-19063. This study demonstrates that a complex containing SAP (synapse-associated protein) 97, CASK, Veli, and Mint1 associates with the C-terminal PDZ-binding domains of Kir2 channels. The authors propose two association models. In one, SAP97 would interact with Kir2 channels and recruit CASK, Veli, and Mint; in the alternative model, Veli would interact with Kir2 channels and and recruit CASK and SAP97.

NHERF-1 uniquely transduces the cAMP signals that inhibit sodium-hydrogen exchange in mouse renal apical membranes
-
Weinman EJ, Steplock D, Shenolikar S. NHERF-1 uniquely transduces the cAMP signals that inhibit sodium-hydrogen exchange in mouse renal apical membranes. FEBS Lett 2003; 536:141-144. This study shows that pharmacological activation of PKA by cAMP inhibits NHE3 activity in BBM isolated from wild-type mice, but has no effect on samples from NHERF1-knockout animals. This lack of response is associated with a reduced cAMP-induced phosphorylation of NHE3.

A kinase-regulated mechanism controls CFTR channel gating by disrupting bivalent PDZ domain interactions
-
Raghuram V, Hormuth H, Foskett JK. A kinase-regulated mechanism controls CFTR channel gating by disrupting bivalent PDZ domain interactions. Proc Natl Acad Sci USA 2003; 100:9620-9625. This paper describes how phosphorylation by PKC of the PDZ2 domain NHERF1 (on Ser-162) decreases the ability of NHERFI to bind CFTR. In addition, a mutation in NHERF1 that mimics phosphorylation (Ser-162 to Asp) decreases CFTR binding and disrupts the ability of NHERF to stimulate CFTR channel activity.

PDZK1: II. An anchoring site for the PKA-binding protein D-AKAP2 in renal proximal tubular cells
-
Gisler SM, Madjdpour C, Bacic D, et al. PDZK1: II. an anchoring site for the PKA-binding protein D-AKAP2 in renal proximal tubular cells. Kidney Int 2003; 64:1746-1754. This paper shows that D-AKAP2, a dual PKA-binding protein, anchors PKA to PDZK1 and to NHERF1.
