Adenosine 5′-triphosphate-sensitive Potassium Channel-mediated Blood-Brain Tumor Barrier Permeability Increase in a Rat Brain Tumor Model
-
NINGARAJ NS, RAO MK, BLACK KL: Adenosine 5′-triphosphate-sensitive potassium channel-mediated blood-brain tumor barrier permeability increase in a rat brain tumor model. Cancer Res. (2003) 63:8899-8911. •• This research article describes a novel cellular mechanism for KATP-mediated BTB permeability increase. This novel mechanism involves the accelerated formation of pinocytotic vesicles, which can transport drugs across the BTB. This article also explains why the KATP channels are a potential target for biochemical modulation of BTB permeability to increase antineoplastic drug delivery selectively to brain tumours. Increased endothelial vesicular transport correlates with increased blood-tumor barrier permeability induced by bradykinin and leukotriene C4
-
The authors of this paper have for the first time shown that bradykinin and leukotriene C4 could increase molecular transport across the BTB. These agents trigger pinocytotic vesicular transport across the BTB, and these transport vesicles may enhance drug transport across the BTB
