Although there is very large number of publications on TPLSM employing FPs as markers, only in few works were the 2PA spectral properties of FPs characterized quantitatively. We tried our best in referring here to all the papers presenting 2PA spectra of different FP mutants in absolute 2PA or 2PE cross section values

The sequence encoding CFP was purchased from Clontech (http://www.clontech.com/), and the one encoding mRFP was a generous gift from Roger Tsien (UC San Diego). These coding regions were moved into the pRSET expression plasmid (Invitrogen; http:// www.invitrogen.com). This produced a His-tagged fusion protein that was enriched first by Ni affinity and then further purified by fast protein liquid chromatography.
