2 + 0.714 mg/ml Probenicid (predissolved in 1 M NaOH) + 0.5 mM brilliant black + 2.5 μM Fluo 4 dye, and incubated at 37.5 °C for 1 h. Ten microliter from compound plates is then added immediately to cell plates using a FLIPR 3 calcium imaging instrument. Fluorescence measurements are then taken. Values given are means of at least three test replicates.
