    • A novel yeast expression system for the overproduction of quality-controlled membrane proteins
    • The authors describe how recombinant membrane protein production is related to the unfolded protein response (UPR) in the yeast S. cerevisiae. They find that some, but not all, membrane proteins elicit a UPR, and that cell-surface expression inversely relates to the degree of the UPR. The best recombinant expression of functional membrane protein is obtained with minimum UPR upon induction.

    • Design of improved membrane protein production experiments: quantitation of the host response
    • The authors quantify the host response to membrane protein production in S. cerevisiae. They find that a tightly defined growth regime is needed for good expression of a recombinant membrane-inserted protein. Differences in membrane protein yield can be attributed to the differential expression of yeast genes involved in membrane protein insertion and in metabolic events, as assessed by microarray experiments.

    • Recognition of transmembrane helices by the endoplasmic reticulum translocon
    • The authors use an in vitro assay to quantify the efficiency with which designed transmembrane (TM) segments insert into dog pancreas rough microsomes. They construct a 'biological' hydrophobicity scale, which surprisingly has much in common with hydrophobicity scales derived from biophysical measurements. This seems to imply that direct protein-lipid interactions are involved in the recognition of TM helices by the translocon. The experiments also reveal a strong dependence on the sequence position of aromatic and charged residues in TM segments.
