ADRB2 polymorphisms and asthma susceptibility: Transmission disequilibrium test and meta-analysis
-
Migita O, Noguchi E, Jian Z, et al. ADRB2 polymorphisms and asthma susceptibility: transmission disequilibrium test and meta-analysis. Int Arch Allergy Immunol 2004; 134:150-157. The authors perform a transmission- disequilibrium test analysis of three polymorphisms in ADRB2 in 137 Japanese families. There is no excess transmission of any of the alleles of the SNPs that they study. The authors then go on and perform a meta-analysis of nine additional association studies for the Arg16Gly and the Glu27Gln SNPs. They do not find an overall association with any of these SNPs and asthma diagnosis.

Meta-analysis of the association of beta2-adrenergic receptor polymorphisms with asthma phenotypes
-
Contopoulos-Ioannidis DG, Manoli EN, Ioannidis JP. Meta-analysis of the association of beta2-adrenergic receptor polymorphisms with asthma phenotypes. J Allergy Clin Immunol 2005; 115:963-972. The authors perform a meta-analysis of the association of Arg16Gly and Glu27Gln with asthma diagnosis and asthma-related phenotypes. They conclude that there is no association between these two coding polymorphisms and either asthma diagnosis of airways hyperresponsiveness. They find an overall association, however, between Gly16 and both nocturnal asthma and asthma severity. Additionally, the authors show differences in allele frequencies between ethnic groups.

Systematic review and metaanalysis of the association between β2-adrenoceptor polymorphisms and asthma: A HuGE review

The authors perform a meta-analysis of the association between Arg16Gly and Glu27Gln with asthma. The authors do not find an overall association with Arg16Gly, but find a weak association with the Glu27 allele. This is the only one of the three meta-analyses that pooled adult studies separately from childhood studies. They also document allele differences between ethnic groups. Significant heterogeneity was found between studies, and interestingly, the heterogeneity persisted when even when only Caucasian studies were included.

Concomitant occasional use of salbutamol influences bronchoprotective responsiveness afforded by formoterol in patients with the glycine-16 genotype

Sims EJ, Lipworth BJ. Concomitant occasional use of salbutamol influences bronchoprotective responsiveness afforded by formoterol in patients with the glycine-16 genotype. Eur J Clin Pharmacol 2004; 59:791-795. This retrospective study found that patients who were both homozygous for the Gly16 allele and were using a long-acting b-agonist were predisposed to greater degrees of airways hyperresponsiveness when they also used short-acting β-agonists.

Use of regularly scheduled albuterol treatment in asthma: Genotype-stratified, randomised, placebo-controlled cross-over trial
-
Israel E, Chinchilli VM, Ford JG, et al. Use of regularly scheduled albuterol treatment in asthma: genotype-stratified, randomised, placebo-controlled cross-over trial. Lancet 2004; 364:1505-1512. This is the first genotype-stratified study of the effects of Arg16Gly on treatment related responses in asthma. Several interesting issues have been raised with regard to this study. In addition to these issues, replication of this study is needed.
