However, their transcriptional role in embryonic stem cell biology is less well understood. Location analysis revealed that Oct4, Sog2 and Nanog are a well-wired transcriptional network that contains highly organized regulatory loops. Furthermore, the coordinative regulation of homeodomain proteins and fate-controlling genes by the network was unexpectedly discovered.

The authors combined the DamID methods with classical mutant analysis, and polytene-staining during early embryogenesis to identify context-dependent Hairy-targets. Hairy was found to be associated with a small group of genes related to the regulation of cell cycle, growth and morphogenesis. Furthermore, this work identifies 'rules' governing recruitment of Hairy co-factors.
