An E. coli strain was constructed harboring an oxidative stress promoter and an SOS-inducible promoter fused to lux and gfp, respectively. The biosensor detected various toxicants with similar sensitivities and intensities to two separate strains containing only one reporter system. The biosensor was able to recognize these toxicants in various mixtures.

Testing of the detection limits and induction factors of these strains towards different genotoxic compounds revealed that the ColD cda promoter was more sensitive and responded more than the more commonly used umuDC, sulA and recA promoters. This finding may contribute to the preferential use of cda in genotox tests in the future.

A Salmonella strain containing a constitutively expressed gfp gene and an SOS promoter fused to lux enabled the detection of genotoxicity and cytotoxicity simultaneously. This study represents a very useful contribution on how to circumvent the problems of non-specific biosensor inhibition leading to false-negative results.

By use of three Salmonella strains, some over-producing nitroreductase and O-acetyltransferase, the usual need for S9 rat liver mixture was avoided and the sensitivity increased. Various mutagenic compounds were detected by the test strains and genotoxicity in extracts of particulate matter from air samples was demonstrated.
