The LucSH cDNA was purchased from InvivoGen (San Diego, CA) and is a fusion between the luciferase and zeocin resistance genes. Both genes have been modified and contain no CpG. The SF91-LUC-EGFP vector co-expressed LucSH and EGFP was pseudotyped with the GALV envelope using the PG13 stable producer line. Tumor cells were transduced and either selected in zeocin or sorted for EGFP+ cells by flow cytometry.
