The most electronegative fraction of LDL from familial hypercholesterolemia patients (named FH-L5) suppresses fibroblast growth factor 2 transcription, resulting in increased apoptosis. PAF-like lipids are suggested to be involved in this apoptotic process. However, the authors failed to isolate an electronegative LDL fraction with the same characteristics as FH-L5 from normolipemic individuals.

Arterioscler Thromb Vasc Biol 2003; 23:1083-1090. The authors succeeded in isolating L5 from normolipemic individuals (N-L5) although in a proportion fourfold lower than FH-L5. However, whereas FH-L5 shows evidence of oxidative modification N-L5 appears as a nonoxidized LDL subfraction. Moreover, FH-L5, but not N-L5, induced endothelial cell-monocyte adhesion, changes in endothelial cell morphology and decreased DNA synthesis and fibroblast growth factor 2 production.

This paper reports that LDL(-) increases tumor necrosis factor α-induced expression of vascular cell adhesion molecule, a process mediated by NF-κB and activating protein 1. This effect is reversed by lipolysis of LDL(-) by lipoprotein lipase. Lipoprotein lipase-treated LDL(-) releases hydroxy-octadecadienoic acids, potent PPARα activators which, in turn, suppress NF-κB and activating protein 1 activation. These findings suggest putative mechanisms that may limit inflammatory responses to LDL(-).
