NADPH oxidase AtrbohD and AtrbohF genes function in ROS-dependent ABA signaling in Arabidopsis
-
The authors show that AtrbohE and AtrbohF NADPH oxidases are required for the production of ROS during ABA-induced stomatal closing in guard cells and during cell expansion in the root. This study provides the first genetic evidence to support a role for ROS in a guard-cell signalling process.

Inducers of plant systemic acquired resistance regulate NPR1 function through redox changes
-
This stimulating paper shows that NPR1 switches from cytoplasmic oligomers to nuclear monomers to activate PR1 expression in response to SA and intracellular redox potential. Mutations of two conserved Cys residues of NPR1 confer constitutive monomer state, nuclear localisation and PR1 activation. These mutations turned SA-dependent oxidative signalling into anti-oxidative signalling, which might explain the relatively slow and indirect nature of the full induction of PR-gene expression by SA.

The Arabidopsis NPR1 disease resistance protein is a novel cofactor that confers redox regulation of DNA binding activity to the basic domain/leucine zipper transcription factor TGA1
-
Després et al. show that redox changes that are influenced by SA control the interaction of NPR1 and TGA1, and hence the DNA-binding activity of TGA1. Further, they show that SA modulates the redox state of TGA1 in vivo through the reduction of two Cys residues that form a disulphide bridge in the absence of SA, and that NPR1 interacts specifically with the reduced form of TGA1. The DNA-binding activity of TGA1 per se is not affected by the redox status; rather, the redox-regulated interaction of TGA1 with NPR1 enhances the DNA binding of TGA1.

Thioredoxin-2 (TRX-2) is an essential gene regulating mitochondria-dependent apoptosis

The work described in this paper suggests that mitochondrial Trx-2 is essential for cell viability. It plays a crucial role in scavenging ROS in mitochondria and in regulating the mitochondrial apoptosis signalling pathway, possibly through the binding of Trx-2 with cytochrome c, a key pro-apoptotic factor.

Transgenic plant cells lacking mitochondrial alternative oxidase have increased susceptibility to mitochondria-dependent and -independent pathways of programmed cell death
-
Robson and Vanlerberghe describe the role that AOX might play in regulating ROS levels in mitochondria in relationship to the induction of PCD. As in animals, cytochrome c is specifically released from mitochondria during ROS-induced PCD in plants. The authors suggest, therefore, that cytochrome c and the induction of AOX may form part of a mechanism that modulates the initiation of a PCD pathway that responds to mitochondrial respiratory status.

Rapid induction of distinct stress responses after the release of singlet oxygen in Arabidopsis
-
The conditional flu mutant of Arabidopsis accumulates the photosensitiser protochlorophyllide in the dark. After a dark-to-light shift, singlet oxygen, a non-radical ROS, is specifically produced. The results described in this article demonstrate that singlet oxygen does not act primarily as a toxin but rather as a signal that activates several signalling pathways. The biological activity of singlet oxygen in Arabidopsis exhibits a high degree of specificity that seems to be derived from the chemical identity of this particular ROS.

Identification of a plant nitric oxide synthase gene involved in hormonal signaling
-
The authors identify an Arabidopsis nitric oxide synthase (Atnos1) mutant that has impaired NO production, resulting in inhibition of organ growth and ABA-induced stomatal movements. This work confirms the importance of NO in crucial processes such as growth and hormone signalling in plants.

Allelopathy and exotic plant invasion: From molecules and genes to species interactions
-
The authors present evidence that Centaurea maculosa, an invasive species in the western United States, displaces native plant species by exuding the phytotoxin (-)-catechin from its roots. Catechin triggers a wave of ROS in the native plant species, which initiates a cascade that triggers changes in gene expression and, ultimately, causes the death of the root system. This work demonstrates another involvement of ROS in death signalling.
