    • The HeLa-Tat-Luc system contains the same reporter plasmid as 5.1 cells and the Tat gene regulated by the CMV promoter. Therefore, the HIV-1 LTR is highly activated in this cell line as a consequence of high levels of intracellular Tat protein.

    • Propidium iodide cell viability: virus infectious and toxicity assays were performed at the same time. Briefly, MT-2 cells were seeded in 24-well microtitre culture plates (5 × 105 per well) and samples were added to obtain a final concentration equal to that used in the recombinant virus assays. After 48 h treatment, cells were collected and analysed by flow cytometry after propidium iodide staining. We determined here the percentage of cells not affected by the sample.
