    • Waterford SD, Kolodecik TR, Thrower EC, Gorelick FS. Vacuolar ATPase regulates zymogen activation in pancreatic acini. J Biol Chem 2005; 280: 5430-5434. The results of this report demonstrate that intracellular digestive enzyme activation, a key pathologic process in acute pancreatitis, is mediated by vacuolar ATPase, which is necessary for creating acidified compartments in the acinar cell. These compartments are the site of digestive enzyme activation.

This report demonstrates that trypsin-mediated activation of PAR-2 has dual effects in the mechanism of pancreatitis. Thus, activated trypsin during pancreatitis mediates both local protective and systemic deleterious responses.

Effects of secretagogues and bile acids on mitochondrial membrane potential of pancreatic acinar cells: comparison of different modes of evaluating DeltaPsim. J Biol Chem 2004; 279:27327-27338. This study shows that secretagogues and bile acids cause depolarization and ATP depletion in pancreatic acinar cells by their effects on intracellular calcium levels. The effects on the mitochondria lead to necrotic cell death.
