Each CoA derivative was synthesized individually by Michael addition of the thiol in CoA to a maleimide-linked fluorescent dye (Molecular Probes, Eugene, OR) and subsequently mixed to prepare dye set A. Fluorescent dyes were chosen in an orthogonal matter, thus eliminating the potential for energy transfer.
