Very interesting chapter that reviews the immunology and genetic factors related to RSV pathogenesis. Nicely dissects the mechanisms of RSV-induced disease in humans and animal models and the role of the innate and the adaptive immune response.

Anti-respiratory syncytial virus (RSV) neutralizing antibody decreases lung inflammation, airway obstruction, and airway hyperresponsiveness in a murine RSV model. Antimicrob Agents Chemother 2004; 48:1811-1822. This study emphasizes the importance of an intervention strategy in RSV disease in the mouse model. The study shows that palivizumab, especially when given before the infection, was associated with improvement of the clinical and inflammatory markers of RSV disease, including long-term AHR and lung inflammation.

    • Bitko V, Musiyenko A, Shulyayeva O, et al. Inhibition of respiratory viruses by nasally administered siRNA. Nat Med 2005; 11:50-55. Very provocative study in which the use of novel siRNA given intranasally before and after RSV infection was able to prevent and inhibit RSV infection in a time-dependent manner. This new agent opens up the possibility for new therapeutic interventions against RSV.
