    • The authors use a CLV3 promoter::β-glucuronidase (GUS) fusion as a marker for stem cell identity to assess the requirement for STM and WUS expression in establishing this cell type. When ectopic WUS activity was induced in the form of a WUS::GR fusion, CLV3::GUS expression was limited to the SAM, whereas when WUS was co-expressed with STM, its expression was also found in leaves. CLV3::GUS expression in STM overexpressers was limited to the shoot apex and to ectopic meristems that formed on leaves.

    • This analysis provides an example of the utility of activation tagging approaches in identifying novel regulatory network components. DÖRNROSCHEN (DRN) is recovered in such a screen and is shown to encode an APELATA2 (AP2)/ETHYLENE RESPONSE FACTOR (ERF)-class protein whose mRNA is expressed in the CZ and at the tips of lateral organs. Plants in which DRN is overexpressed undergo SAM arrest, form radialised leafs, and show altered patterns of STM, CLV3 and WUS expression. No loss-of-function phenotype was seen for DRN, most likely because of redundantly acting genes. A role for DRN in repressing stem cell fate is hypothesized on the basis of expression and overexpression phenotypes.
