    • Vercelli D. Genetics, epigenetics, and the environment: switching, buffering, releasing. J Allergy Clin Immunol 2004; 113:381-386. This article reviews the emerging relationship between genes and environment in the pathogenesis of complex diseases, such as asthma. The article examines this interaction by using CD14 as a model. Fully understanding gene-environment interaction requires that epigenetic as well as classical genetic mechanisms be taken into account.

    • Jones MG, Nielsen J, Welch J, et al. Association of HLA-DQ5 and HLA-DR1 with sensitisation to organic acid anhydrides. Clin Exp Allergy 2004; 34:812-816. This study included 52 cases and 73 referents. They found that within DQ5, DQB1*0501 confers susceptibility to develop sensitization to organic acid anhydrides. Interestingly, DQB1*0501 is protective for LMWAs such as isocyanates and plicatic acid, which may indicate varying affinities for the corresponding specific Class II molecules. However, at variance with anhydrides, an IgE mechanism is unlikely to occur in isocyanate and plicatic acid-induced asthma.

HLA-DR7 was associated with both sensitization and respiratory symptoms at work, whereas HLA-DR3 was protective against sensitization.

J Allergy Clin Immunol 2003; 111:1087-1094. In this HDI murine model, mice were epicutaneously exposed to HDI and then challenged with HDI by means of inhalation to induce airway inflammation or on the ear to induce contact hypersensitivity (CHS). In contrast to CHS, induction of Th2 responses after epicutaneous exposure is strongly influenced by genetics. The lung inflammatory response to inhaled HDI depends primarily on effective generation of CD4(+) Th2 responses, as in the case of atopic asthma.

The lack of TNF-α inhibited the development of allergic rhinitis as shown in mice deficient in the gene encoding TNF-α.
