    • Formation of dendritic spines with GABAergic synapses induced by whisker stimulation in adult mice
    • The authors show that 24 h of single whisker stimulation in adult rats increases the density of GABAergic synapses on spines but not dendritic shafts in layer IV of the corresponding barrel. This morphological change is accompanied by altered responses to peripheral stimulation. Structural and physiological changes are still present four days after stimulation, demonstrating that activity can induce long-lasting changes in a specific cortical inhibitory circuit.

    • Dark rearing alters the development of GABAergic transmission in visual cortex
    • The authors demonstrate that dark rearing prevents the postnatal increase of inhibitory inputs onto layer II/II neurons in rat visual cortex. Visual deprivation by dark rearing prevents the developmental increase in the number of release sites but increases the strength of single GABAergic inputs. The authors suggest that sensory experience plays a permissive role in the development of cortical inhibition by increasing the strength of preexisting GABAergic connections.

    • Coincident pre- and postsynaptic activity modifies GABAergic synapses by postsynaptic changes in Cl- transporter activity
    • This study demonstrates spike time dependent plasticity in hippocampal GABAergic synapses. Coincident pre- and postsynaptic activity induces a long-term decrease in the activity of a potassium-chloride cotransporter, which in turn increases the postsynaptic intracellular chloride concentration. Depending on the initial value of the chloride equilibrium potential this shift can decrease or increase GABAergic postsynaptic currents.

    • Experience-dependent refinement of inhibitory inputs to auditory coincidence-detector neurons
    • Using a variety of immmunohistochemical markers the authors demonstrate a dramatic developmental redistribution of glycinergic synapses in neurons in the medial superior olive of gerbils. Before hearing onset, many glycinergic synapses contact proximal dendrites but after two weeks of normal hearing dendritic synapses have become eliminated. By contrast, somatically located glycinergic synapses are not eliminated. Rearing gerbils in omnidirectional noise significantly increases the density of dendritic inhibitory synapses. Unilateral cochlear ablation before hearing onset results in a dramatic increase of glycinergic synapses both on the soma and dendrites and a uniform subcellular distribution of inhibitory synapses.

    • Elimination and strengthening of glycinergic/GABAergic connections during tonotopic map formation
    • This study uses focal uncaging of caged glutamate to demonstrate that functional topography in the GABAergic/glycinergic MNTB-LSO pathway of rats is substantially refined during the first postnatal week. Minimal stimulation techniques further reveal a fourfold decrease in the number of MNTB neurons converging on single LSO neurons and a 12-fold increase in the synaptic conductance of maintained individual MNTB-LSO connections. This study provides the first direct evidence for specific functional elimination of GABAergic/glycinergic synapses during establishment of a defined inhibitory circuit.

    • This study shows that GABAergic/glycinergic synapses in the MNTB-LSO pathway elicit an increase in the intracellular calcium concentration. Together with Kullmann and Kandler [43], this study demonstrates that depolarizing GABAergic/glycinergic synapses in the LSO are indeed excitatory during the period when most of these synapses are functionally eliminated.

    • An instructive role for retinal waves in the development of retinogeniculate connectivity
    • This study is an elegant demonstration that neuronal activity plays an instructive role in segregation of eye-specific layers in the lateral geniculate nucleus. Instead of decreasing neuronal activity, the classical way neuronal activity is altered, the authors pharmacologically increase the intracellular levels of cAMP in the developing retina in vivo, which increases the frequency of retinal waves in the treated eye relative to the untreated eye. Consistent with an introductive role of neuronal activity, retinal ganglion fibers from the more active eye take over territory from the less active eye and occupy larger areas in the lateral geniculate nucleus.

    • Deafness disrupts chloride transporter function and inhibitory synaptic transmission
    • The authors show that bilateral cochlear ablation performed about one week before hearing onset reduces the amplitude of GABAergic postsynaptic potentials in the inferior colliculus when tested around hearing onset. Weakening of inhibitory postsynaptic potentials is presumably caused by a decrease in the activity, but not the expression level, of the potassium-chloride cotransporter KCC2. As these changes are induced before hearing onset, integrity of the auditory periphery and/or normal spontaneous activity patterns are required to maintain normal chloride homeostasis in auditory midbrain neurons.

    • The authors combine electrophysiological and electron microscopic techniques to demonstrate that the switch from a mixed GABA/glycinergic MNTB-LSO pathway to a pure glycinergic MNTB-LSO pathway [52] is regulated on a single cell level.
