    • Ephrin-A5 (AL-1/RAGS) is essential for proper retinal axon guidance and topographic mapping in the mammalian visual system
    • This paper reports the first knockout of an ephrin ligand, ephrin-A5, and describes defects in retinal axon guidance and topographic mapping in ephrin-A5-deficient mice. The authors show that ephrin-A5 is required for proper topographic mapping of retinal axons to the superior colliculus, and, in addition, it appears to act as a repellent barrier restricting retinal axons to the superior colliculus

    • Eph receptors discriminate specific ligand oligomers to determine alternative signaling complexes, attachment, and assembly responses
    • Reports the remarkable finding that although an artificially dimerised soluble ephrin can stimulate Eph receptor phosphorylation, a cell attachment response only occurs after activation with higher-order ephrin multimers. This correlates with the recruitment of a phosphotyrosine phosphatase to the receptor. These findings raise a number of possibilities regarding modulation of receptor function and adhesion versus repulsion responses
