Interestingly, the zygote expresses two WOX genes, which are subsequently active in either of the zygote derivatives. Furthermore, WOX expression patterns define novel cell fates that are not morphologically distinct. Analysis of the wox2 mutant shows that WOX proteins may also contribute to the specification of the cells that express them.

This paper describes the isolation of three mutations that specifically affect suspensor development. YODA encodes a MAPK kinase kinase, and loss-of-function alleles impair specification of the basal cell as suspensor. Gain-of-function alleles cause the converse phenotype and convert embryo cells to suspensor-like fates.

Local auxin accumulation depends on auxin transport activity. Using detailed expression analysis, the authors show that four PIN proteins are expressed in the embryo, and that their subcellular localization corresponds to the distribution patterns of auxin. Finally, a quadruple mutant in PIN1, PIN3, PIN4 and PIN7 shows a dramatic embryo phenotype. Taken together, the results described in this paper provide a framework understanding of auxin action during embryo development.

Results described in this paper, together with subcellular localization of GN to endosomes, provide convincing evidence that GN regulates auxin efflux by promoting the recycling of PIN1 from endosomes to the plasma membrane.
