This study, together with previous studies from the same laboratory, describes an interesting mechanism of dual targeting of the cytochrome P450 2E1 (CYP2E1) to the ER and mitochondria. The authors show that CYP2E1 presents a cryptic mitochondrial presequence, in addition to the previously demonstrated ER signal sequence, within the first 30 amino acids. cAMP-mediated phosphorylation of Ser-129 increases the mitochondrial targeting of the protein but does not affect the translocation of CYP2E1 to the ER.

In a wide survey of the nucleocytoplasmic partitioning of the plant photoreceptors phytochromes A, B, C, D, and E, the authors show that all five phytochromes are imported into the nuclei and that this process is differentially regulated by light. In addition, the authors show that this nucleocytoplasmic partitioning is also regulated by diurnal rhythm, suggesting that it has a role in specifying the functions of these photoreceptors.

Alternative translation initiation of a single mRNA carrying two in-frame start codons is involved in the subcellular localization of dual-targeted proteins in the mitochondria and chloroplasts. In this study, the authors showed that a less favorable context for translation initiation at the second AUG is used to direct the translocation of THI1 to mitochondria. The subcellular localization of THI1 could involve a cap-dependent translational mechanism around the first AUG and a cap-independent translation process at the second start codon.

The authors describe their careful study on the cellular and subcellular localization of phototropin 1 in various organs and cells of Arabidopsis, in which they used a phot1::green fluorescent protein (GFP) fusion protein as a reporter. They showed that this photoreceptor moves from the plasma membrane to the cytosol in response to blue light, which could have a direct effect on signaling pathways.

The authors investigated the domain structures of the glutathione reductase presequence for the first time. This enzyme is one of the proteins that carries an ambiguous targeting sequence that allows it to be dual-targeted to mitochondria and chloroplasts. The authors were able to identify distinct domains within the signal peptide that control the efficiency of import into both mitochondria and chloroplasts.

The authors demonstrate that the transport of malfolded proteins in the secretory pathway, after retrograde exit from the ER, is cytoskeleton-dependent and finally results in their accumulation within the nucleoplasm. This interesting work is not a typical example of mutual targeting as it is more related to a quality control mechanism, but it clearly shows that a single polypeptide can accumulate into different subcellular compartments despite the absence of a particular targeting signal.
