This study reports the three-dimensional structure of mammalian Sar1p bound to GDP. The authors use this information to guide a mutation analysis of Sar1p. The data suggest that distinct amino-terminal regions of Sar1p are involved in nucleotide exchange and hydrolysis. Although many questions remain on Sar1p function vis-à-vis structure, this report represents a milestone in studies on the COPII budding machinery.

Rotary shadowing and electron microscopy are used to visualize individual Sec23p-Sec24p and Sec13p-Sec31p complexes as well as COPII coated vesicles. The surface of these vesicles consists of repeating polygonal units with side lengths of these units close to the dimensions of a rod-like domain observed in Sec13p-Sec31p. Additional biochemical studies indicate the Sar1p and Sec23p-Sec24p subunits are within 1 nm of the phospholipid head groups of a coated bilayer whereas the Sec13p-Sec31p subunit is displaced at least 1.6 nm from the bilayer surface.

Moreover, a single carboxy-terminal valine reside in the -1 position can act as an efficient export signal. The authors note that these motifs are common among other type I membrane proteins that are transported from the ER.

A strong case for the receptor-mediated export model is provided for Erv29p-dependent packing of glyco-pro-alpha factor into COPII vesicles. In vitro experimentation indicates a direct interaction between Erv29p and glyco-pro-alpha factor, and in vivo studies suggest a saturatable relationship between this receptor and its ligand. Although the study was conducted in yeast cells, Erv29p is found in all species examined, suggesting a general mechanism.

Following the degradation of misfolded secretory proteins such as CPY*, the authors report that anterograde transport between the ER and Golgi is required for efficient turnover. Furthermore, other proteins that operate in transport through the early secretory pathway influence CPY* degradation rates, including Erv29p. The report is the first to document that several soluble secretory proteins accumulate in the ER of erv29 deletion strains.

Monitoring a variety of misfolded secretory proteins, this report shows that some of these substrates depend on transport between the ER and Golgi compartments for degradation. It is proposed that certain misfolded proteins depend on anterograde transport to the Golgi complex and then retrograde transport back to the ER for degradation. Although similar conclusions are reached as in Caldwell et al. (2001) [39•], further tests may be necessary to exclude indirect consequences of these transport blocks on ER quality control.
