Plant Physiol 2002, 129:1507-1520. The authors co-expressed dominant negative forms of the small GTP-binding proteins ARF1 and ARF3 with FP markers for the Golgi and plasma membranes. ARF1 but not ARF3 function was required for the intracellular trafficking of both markers. Redistribution of the Golgi marker to the ER revealed an altered ER morphology when the dominant negative ARF1 was expressed, suggesting that ARF1 function is also required for maintenance of normal ER morphology.

Plant Cell 2003, 15:1057-1070. The expression of a dominant negative mutant of the small GTP-binding protein Rha1 causes two FP fusions destined for the vacuole to relocalize. The redistributed protein co-localizes with a marker for the prevacuolar compartment, suggesting that Rha1 plays a role in trafficking between the prevacuolar compartment and the vacuole. This role appears to be specific to Rha1, as expression of a dominant negative form of another Rab5 homolog, Ara6, had no effect on the distribution of these markers. A nice example of the use of red fluorescent protein (RFP) and GFP co-expression.

Plant J 2002, 31:499-515. The authors analyze three FP markers for the Golgi apparatus and one marker for the vacuolar lumen in tissue culture cells expressing GTP-or GDP-fixed mutations in ARF1. Their findings suggest that ARF1 is required both for trafficking between the ER and Golgi and for maintenance of Golgi organization.

The measurement of individual microtubule dynamics using GFP::tubulin fusions and photobleaching experiments provides evidence of the cortical sites of origin for interphase microtubules, for dynamic minus ends, and for the widespread motility of cortical microtubules by a treadmilling mechanism. Treadmilling motility was observed to contribute to the formation of microtubule bundles.

The authors suggest that the regulation of catastrophe rate plays a role in the formation of the pre-prophase band.

Interphase chromosomal dynamics are explored by marking discrete chromosomal sites with Lacl::GFP bound to tandem arrays of Lacl-binding sites. The tagged domains show a greater range of movement relative to the DNA concentration in the nuclei of polyploid pavement cells than in the diploid nuclei of neighboring guard cells. These results suggest that chromatin is relatively less constrained in the endoreduplicated nuclei.

These fusions suggest that KAPP promotes the internalization of SERK1 from the plasma membrane. SERK1 and KAPP co-localize at the plasma membrane and on internalized vesicles, but FRET and FLIM analyses indicate that these proteins interact primarily in the internalized vesicles. An excellent example of the use of FRET to distinguish between co-localization and interaction.

A fluorescent protein-based voltage sensor is improved to produce both variants that are suitable for the measurement of rapid voltage changes, such as action potentials, and variants that can be imaged ratiometrically. Of special interest is the fact that the changes in emission properties of these voltage sensors appear to be caused by a conformation change in the fluorophore, which is conferred by the fused channel protein.
