E and consequently the response to misfolded proteins in the cell envelope. Sequential degradation of RseA by the membrane-anchored DegS protease and the membrane-embedded metalloprotease YaeL is demonstrated. Thus, release of an initially membrane-sequestered transcription factor by a proteolytic event is a mechanism of gene activation, which conceptually and, even in part mechanistically (YaeL is a homolog of the mammalian S2P protease), is similar in prokaryotic and eukaryotic cells.

In the absence of AAI, TraR is rapidly degraded by Clp and Lon proteases. This proteolytic control should contribute to the switch-like induction of TraR as well as to termination of the quorum-sensing response above or below a certain AAI threshold concentration.

The proteolysis-promoting 11-amino-acid SsrA tag is shown to provide contiguous binding sites for the cooperating factors ClpX and SspB, whereas the binding site for ClpA overlaps with that for SspB (each site consisting of three to five amino acids only). Besides demonstrating high informational content of the short SsrA tag, these findings explain the physiological interference in protein recognition between SspB and ClpA.

Science 2001, 293:705-708. Polyphosphate accumulates in cells exposed to different kinds of starvation. It is shown that polyphosphate specifically binds to Lon protease, which directs Lon towards recognition and degradation of excess ribosomal proteins. Thus, polyphosphate contributes to the mobilization of intracellular amino acid resources in starving cells.
