Proc Natl Acad Sci USA 2001, 98:14895-14900. The role of IscR in transcriptional repression of the E. coli isc operon is documented. The finding that IscR is a [2Fe-2S] cluster protein and that Iscr is not functional in strains lacking the Fe-S cluster assembly protein, IscS, suggests a novel feedback loop for regulation of Fe-S cluster assembly.

Examination of the properties of the two E. coli aconitases indicates that AcnA is resistant to superoxide in vivo because there is a factor present in cells that normally protects this activity from oxidative damage. In contrast, AcnB activity is not associated with a protective factor and its cluster is labile under normal growth conditions. Loss of AcnB activity also causes the excretion of citrate, an iron chelator for which E. coli synthesizes a transport system, which indicates a connection to iron metabolism.

In this study, soda expression is shown to be affected by the levels of aconitase, presumably at the level of translation. In addition, strains lacking aconitase show changes in the levels of several proteins and are sensitive to redox agents, reinforcing the notion that aconitase has a global function.
