The finding that IscR is a [2Fe-2S] cluster protein and that Iscr is not functional in strains lacking the Fe-S cluster assembly protein, IscS, suggests a novel feedback loop for regulation of Fe-S cluster assembly.

Examination of the properties of the two E. coli aconitases indicates that AcnA is resistant to superoxide in vivo because there is a factor present in cells that normally protects this activity from oxidative damage. In contrast, AcnB activity is not associated with a protective factor and its cluster is labile under normal growth conditions. Loss of AcnB activity also causes the excretion of citrate, an iron chelator for which E. coli synthesizes a transport system, which indicates a connection to iron metabolism.
