    • The use of scFvs as intrabodies to block their target's function is often limited by the sensitivity of scFvs to the reducing environment of the cytosol, and specific methods have been developed to overcome this limitation (24-26). Accordingly, the scFvs we have selected from the Griffin. 1 library so far, and have expressed intracellularly, did not inhibit their target function. However, a large proportion of these scFvs efficiently detects their endogenous target upon moderate expression and can in turn be used to track endogenous protein dynamics. Whether this unexpected high frequency of in vivo active scFvs is due to our expression conditions, or to the presence of many reduction-resistant scaffolds in the Griffin. 1 library, is at the moment unknown.
